Resumen:
Adenosine A1 receptor agonist induces gene expression changes in light induced retinal degenerationSoliño M1,Larrayoz I2, López EM1, Rey-Funés M1, Martinez A2, Girardi E1y López-Costa JJ1.A1 receptor (A1R) has been described in the retina and has been linked to the control of the characteristics of the subretinal space. A1R plays a role in inflammation, neuronal and trophic factor activity. Recently at our lab, the modulation of A1R emerged as an effectiveneuroprotective strategy of the retina in the model of light induced retinal degeneration (LIRD). In order to further characterize the neuroprotective effectsunderlying A1R agonist treatment,mRNA changes induced by N6-cyclopentyladenosine (CPA), an A1R agonist, were studied in theretina using aLIRD model.Sprague Dawley rats received a single intravitreal injection of either CPA orvehicle ,and then were submitted to continuous illumination (CI) (12000 lux) during 1 day. After this, theretinas were extracted and processed by RT-PCR. We selected as genes of interest adenosine receptors (A1, A2a, A2b and A3), proangiogenic factors (adrenomedullin and VEGF), a trophic factor (PEDGF), inflammation mediators (IL-1b, iNOS, nNOS and TNF) and a glial reactivity marker (GFAP).Data werestatistically analysed using t-teston the Graphpad Prism software.RT-PCR results showeda significant decrease of A1, A2a and A2b receptors mRNA in the retinaof CPA treated eyes (p
