CB2 RECEPTOR EXPRESSION AND ENDOCANNABINOID ENZYMES IN RAT RETINA AND ITS MODIFICATIONS AFTER CONTINUOUS ILLUMINATION

The endocannabinoid (eCB) system plays a neuromodulatory role in the retina and it could be a potential therapeutic target in light induced retinal degeneration (LIRD). Thence the aim of this work was to study the expression of cannabinoid receptor-2 (CB2) and of the eCB system related enzymes in control and CI exposed rats.Sprague Dawley rats were submitted to regular illumination (CTL, 80 lux, light /dark cycles 12:12hs) or continuous illumination (CI) (12000 lux) during 1, 2, 5 and 7 days. The eyes of the rats were processed by immunocytochemistry (ICC) or western blot (WB) using a primary antibody to CB2 (Abcam). RT-qPCR was employed to quantify mRNA of CB2, FAAH, and COX2 enzymes.After 1 day of CI, CB2 levels increased detected by WB. At 5 days of CI, a greater rise from CTL levels was detected by WB and also by RT-qPCR. After 7 days of CI, CB2 levels were still higher determined by both techniques. ICC showed particular changes in different retinal layers. Interestingly, CB2 was upregulated at days 1 and 2 of CI in the photoreceptor layer. Among the studied eCB enzymes, FAAH mRNA significantly diminished along the 7 days of CI, while on the opposite, COX2 mRNA increased in the same period.The observed plastic changes in the eCB system are consistent with a potential role of this system in the LIRD model. The rise of CB2 expression at 24hs coexists with the previously determined maximum oxidative damage offering a suitable therapeutic window in the model.
