Various models have been developed to describe the mechanisms responsible for the fluctuations in the ligand-activated ionic channels of cell membranes. However, no single model seems capable of accounting for all the complexity exhibited by these channels. Here we propose a new formulation that integrates features of older models, and lends itself to a direct physical interpretation. Starting from a generalization of the defect-diffusion model originally proposed by Millhauser and Läuger, we have included the two-gate desensitization scheme introduced by Auerbach. This procedure allows us to consider simultaneously such features as channel desensitization and the dependence on agonist concentration. We solve the model is solved and analyze and interpret its predictions, comparing them with experimental results.
