P1 Epigenetic Regulation in Leaves of High Altitude Maize Landraces:Effect of UV-B Radiation

P1isaR2R3-MYBtranscriptionfactorthatregulatestheaccumulationofaspecificgroupofflavonoidsinmaizefloraltissues,suchasflavonesandphlobaphenes.P1isalsohighlyexpressedinleavesofmaizelandracesadaptedtohighaltitudesandhigherlevelsofUV-Bradiation.Inthiswork,weanalyzedtheepigeneticregulationoftheP1genebyUV-Binleavesofdifferentmaizelandraces.OurresultsdemonstratethatDNAmethylationintheP1proximalpromoter,intron1andintron2isdecreasedbyUV-Binalllinesanalyzed;however,thebasalDNAmethylationlevelsarelowerinthelandracesthaninB73,alowaltitudeinbredline.DNAdemethylationbyUV-BisaccompaniedbyadecreaseinH3methylationatLys9and27,andbyanincreaseinH3acetylation.smRNAscomplementarytospecificregionsoftheproximalpromoterandofintron23′endarealsodecreasedbyUV-B;interestingly,P1smRNAlevelsarelowerinthelandracesthaninB73bothundercontrolconditionsandafterUV-Bexposure,suggestingthatsmRNAsregulateP1expressionbyUV-Binmaizeleaves.Finally,weinvestigatedifdifferentP1targetsinflowertissuesarealsoregulatedbythistranscriptionfactorinresponsetoUV-B.SometargetsanalyzedshowaninductioninmaizelandracesinresponsetoUV-B,withhigherbasalexpressionlevelsinthelandracesthaninB73;however,notallthetranscriptsanalyzedwerefoundtoberegulatedbyUV-Binleaves.
