VARIACIONES EN LA ESTIMACION DE LA ABUNDANCIA ABSOLUTA

RESUMEN Con el objeto de estimar las variaciones numéricas de distintos componentes del picoplancton en relación con el tiempo transcurrido entre la colecta-preservación de la muestra y su procesamiento (tinción con DAPI), y teniendo en cuenta dos áreas hidrológicamente disímiles (Mar Argentino y Mar de Weddell), se examinaron dos puntos espaciales durante el verano 2002-2003. Durante el tiempo que duró la experiencia se observó para ambos sitios una pérdida celular constante que promedió el 1% diario, siendo algo mayor para la muestra proveniente del Mar Argentino. Esta tasa concuerda con el valor reportado en la literatura (39% en 40 días; Turley, 1993) e indica firmemente que, independientemente del área bajo estudio, la abundancia del picoplancton sólo puede ser evaluada con un grado aceptable de precisión mediante el análisis de muestras procesadas en forma inmediata luego de su fijación.   ABSTRACT Two hydrologically dissimilar locations (Argentine Sea and Weddell Sea) were sampled during austral Summer 2002-2003 with the aim of estimating quantitative variations of different picoplanktonic components as related to the time elapsed between collection/fixation of the sample and its further processing with DAPI. A constant cell loss of approx. 1% per day was observed during the time of the study for both sites, although slightly higher for the sample from the Argentine Sea. Such a rate is in agreement with the value reported in literature (39% in 40 days; Turley, 1993), thus firmly indicating that, regardless of the area being studied, picoplanktonic abundance can only be assessed with an acceptable degree of precision through the examination of samples that are processed immediately after their fixation.
