Soybeans (Glycine max) is an important crop and the main source both of oil and protein through the world. At present there is little information on the natural occurrence of Fusarium toxins in this substrate and by-products. Deoxynivalenol (DON) and nivalenol (NIV) are frequent contaminants of cereal and oil seeds worldwide, these toxins have adverse effects on animal and human health. The aim of this work is to develop a method for DON and NIV determination in soybean seeds and flour. The toxins were extracted with acetonitrile-water (84:16), cleaned up on a Romer MycosepTM 225 column, detected and quantified by high performance liquid chromatography (HPLC). The in house-method characteristics of linearity, accuracy, presicion and detection/quantification limits were defined by means of recovery test with spiked soybean and soybean flour samples in a range of 1-8 µg/g for both toxins. Recoveries ranged from 75 to 92% for DON and 70 to 90% for NIV with relative standard desviation generally lower than 9 % for DON and 12% for NIV. The detection limit of the method was 0,1 µg/g for DON and 0,2 µg/g for NIV, based on a signal -to-noise ratio 3:1
