Regulatory T cells (Treg) accumulate inside tumors: understanding which signals regulate their function is crucial. Here, we investigate the effect of type I IFN (IFN-I) on Treg biology. We developed a new strain of mice (IFO) by backcrossing Foxp3GFP females with IFNAR-/- males lacking the IFNAR1 subunit of the IFN-I receptor. After the fourth backcross cycle an homozygous strain was obtained. The leukocyte population in spleen, inguinal and mesenteric lymph nodes, thymus and blood in IFO animals showed a similar pattern to IFNAR-/- mice assessed by Flow cytometry, with reduced numbers of NK, CD4+, CD4+Foxp3+, CD8+ cells in spleen and MLNs compared to Foxp3GFP mice (p
