Enhancement of immunological activity of CpG-ODN vehiculized in lyotropic liquid crystal carriers

CpG-ODN (oligonucleotides containing unmethylated CpG inparticular base context) is a new adjuvant for vaccines thatunfortunately has shown reduced bioavailability owed mainly tolack of chemical stability. In this work, we evaluated theadjuvant capacity of CpG-ODN incorporated into lamellarmesophases of lyotropic liquid crystals (coagels). For that,BALB/c mices were immunized (s.c.) three times during fifteendays (schedule: 0, 7 and 15) with a protein antigen (OVA, 60ug/mice/dose) plus CpG-ODN (50ug/mice/dose) or OVA/CpGODN/coagel. On day 21, we measured specific antibodies inplasma and specific cytokines in cell culture supernatants(ELISA). The measured titles (log10) were:IgG IgG1 IgG2aOVA/CpG-ODN 3,9±0,0 3,4±0,1 2,7±0,2OVA/CpG-ODN/coagel 5,1±0,0 4,4±0,1 3,8±0,2The levels of IFN- were higher in animals immunized withOVA/CpG-ODN/coagel than in those immunized withOVA/CpG-ODN (p<0,005). These results indicate that CpGODNintensified the specific immune response whenadministered in coagels.
