Brasil   The basidiomycetous fungus Cryptococcus gattii causes respiratory and disseminaed infections that affect predominantly immunocompetent individuals. This microorganism is a facultative intracelular pathogen that can use host macrophages to disseminated more efficienlly, evade the host immune response, and under certain circumstances survive in a dormant state. The aim was identify C. gttii genes differentially induced during macrophages infection. The differences between the two cDNA population were analyzed by representational difference analysis (RDA).RDA was performed using cDNA from C. gattii grow within rat peritoneal macrophages tor 24 h (TESTER) and eDNA Irom C. gattii grow in RPMI1640 tor 24 h (DRIVER). Final RDA products generated after two rounds of amplification were cloned in pUCI8. Plasmidial DNA was prepared from 200 clones and sequenced. The resulting sequences are being compared to the Genbank database using the tBLASTX program. CONCLUSION: The identification of genes expressed during macrophages infection can be exploited for the development of novel prevention and treatment strategies for cryptococcosis.
