Resumen:
Epidemiological data suggest that chronic exposure to Chlorpyrifos (CPF), one of the most used pesticides, leads to increased risk of pregnancy disorders. Here, we assessed the effects of CPF on villous cytotrophoblasts (CTBs) and explants from human normal-term placentas. This study was approved by the Hospital Privado Ethics Committee, Argentina. Morphology, cell viability and gene expression were analyzed in samples exposed to increasing CPF concentrations up to 100µM for up to 64h. In these conditions, CTBs differentiated to syncytium-like structures as the non-exposed control; showed higher expression of Pg-p, ABCG2 and βhCG; and did not present signs of cellular dead. However, morphological alterations and cellular dead signs were observed in placental villous explants exposed to the same CPF concentrations. These results support and extend our previous findings obtained in JEG-3 cells and suggest that CPF cytotoxicity on trophoblast may be influenced by surrounding components of the placental villi. * Ph.D Students
