Resumen:
Objectives: a) to study the expression of hyaluronan (HA) andRHAMM, b) toevaluate the mRNA expression of HAS2-3 and Hyals1-2 in choriocarcinoma(CC) cell lines, c) to determine the effect of different concentrations (62.5 to500 mg/ml) of several molecular weight HA (high-HMW-, low-LMW- andoligomers-o-) in migration and proliferation, d) to analyze the expression ofRHAMM and signaling pathways involved in the JEG-3 cell line.Methods: human CC cell lines JAR, JEG-3 and BeWo were used. HAsecretion was measured by an ELISA-like assay. mRNA and proteinexpression were evaluated by RT-PCR, western blot and immunofluorescence,respectively. Cell proliferation was studied by [3H]-thymidine uptakefor 48 and 72 h. Migration was assessed by wound healing andtranswell assays for 24 h. Ly294002 (PI3K inhibitor) and UO126 (MEK1/2inhibitor) were used to evaluate signaling pathways.Results: All CC cell lines showed expression of HAS and Hyals while onlyBeWo showed higher basal secretion of HA respect to its cell-free medium(p
