Decoction of roots and rhizomes of this plant are used as diuretic, diaphoretic and antirheumatic folk medicine. A bone-protective role of several plant polyphenols, such as flavonoids which are present in SM, was found in menopause and rheumatoid arthritis. The aim of the study was to determine whether SM aqueous extracts inhibit RANK ligand (RL)-elicited osteoclast differentiation of macrophages, a molecular mechanism induced during rheumatoid arthritis. RAW 264.7 murine macrophages were cultured during 5 days with RL (100 ng/ml) in the presence or absence of SM (1-10-100-1000 ng/ml). We measured: multinucleated, tartrate resistant acid phosphatase (TRAP) positive (osteoclast-like) cells, by cytochemistry; cathepsin K mRNA expression, by RT-PCR; matrix metalloproteinases (MMP) activity, by gel-zymography; and cell proliferation, by BrdU incorporation; in order to asses osteoclast differentiation of RAW cells. Treatment of RAW cells with SM diminished RL-induced osteoclast-like cells (OLCs), cathepsin K mRNA expression and MMP activity, in a dose dependent manner. Significative inhibition (p<0,05) of OLCs (30±10%), and MMP activity (37±16%) was found with SM 100 ng/ml. Higher concentration of SM (1000 ng/ml) was needed to reach a significative inhibition (44±20%) of cathepsin K mRNA expression. The inhibition found was not due to a cytotoxic effect exerted by the extract, since SM (1-1000 ng/ml) did not affect cell proliferation of RAW cells, nor the increase of cell proliferation induced by RL. We conclude that aqueous extracts of SM are capable to significantly diminish osteoclast differentiation of RAW macrophages induced by RL in vitro. This mechanism could be involved in the therapeutic role of zarzaparrilla plant as antirheumatic folk medicine.
