In the drug delivery field, the development of new materials may achieve a controlled release of biactives. In this study, we performed an in vivo evaluation of chitosan hydrogels modified and unmodified with DDSA (dodecenyl succinic anhydride), loaded with thymol, an antioxidant-antimicrobial compound which may help to treat the harmful effects of chronic inflammation developed in the periodontal disease. Results showed that the modification of chitosan with DDSA influenced the drug delivery profile and in vivo performance of the hydrogels. In vitro thymol release from DDSA-chitosan hydrogels lasted almost seven days whereas from chitosan hydrogels it took only three days. Nevertheless, histology evaluation revealed that, in a rat periodontitis model, with the local application of the hydrogels every two days for a week, only the chitosan hydrogels loaded with thymol seem showed lower sings of periodontal tissue damage.
