Stevia rebaudiana bertoni is an herbaceous plant widely distributedand used for its sweetener character. In previous studies carried out inour laboratory, we demonstrated that oral administration of stevioside(S), a mayor component of stevia, improved the recovery of contractileactivity in hearts subjected to I-R and decreased the infarct size.These effects were, at least in part, reverted by the administrationof Wortmannin (W), an inhibitor of Akt. The aim of the present studywas to investigate the mitochondrial morphology and ATP synthesiscapacity after oral administration of S (168 mg/kg for 15 days) andits relation with the activation profile of Akt and GSK3β, in Langendorffperfused rat hearts subjected to I-R. Hearts from female Wistarrats (200-250g) fed ad libitum were used. W (100nM) was added 15min before I. Mitochondrial ultrastructure was analyzed by electronmicroscopy, the measurement of mitochondrial ATP synthesis wasmade by the luciferin-luciferase method and the activation profileof Akt and GSK3β were studied by western blot considering Akt-P/Akt-T and GSK3β-P/GSK3β-T, respectively.
