Effects of erythropoietin on heart function and oxidative damage in age-associated hypovolaemic conditions

Introduction: When severe hypovolaemic state occurs, mortality increases in old human compared with young. Aging increases perioperative complications of cardiovascular surgery. Cardioprotective function of erythropoietin (EPO) has been extensively studied. However, the role of EPO on cardiovascular system after haemorrhage and its effect during aging process is unclear. Purpose: we examined the effects of EPO pre-treatment on cardiac function and oxidative damage following haemorrhage 20% of the volaemia (H) during advance of age. Methods: male Sprague-Dawley aged rats 2 (young: Y) and 18 (adult: A) months old were treated with either 1 ml/kg PBS (phosphate-buffered saline) or 1000 IU/kg EPO i.p daily for 3 days pre-treatment. Groups of rats: (n=8 each group): S: sham + PBS pre-treatment; E: sham + EPO pre-treatment; H: H+PBS pre-treatment and H+EPO: H+EPO pre-treatment. On day 4, rats were subjected to surgical procedure only or haemorrhage. Mean arterial pressure (MAP, mmHg) and heart rate (HR, bpm) were evaluated during 120 min. At the end of each experiment we determined: echocardiographic parameters, serum EPO concentration and haematocrit. Thiobarbituric acid-reactive substances (TBARS) were evaluated in left ventricle tissue homogenates.
