Antiproliferative and antioxidant activities of Tilia x viridis: contribution of monoterpenes and scopoletin

  In previous studies a dichloromethane extract (DME) from Tilia cordata presented selective antiproliferative action on a lymphoma cell line (BW5147) [1]. Hydrogen peroxide (H2O2) and O2- are related with the modulation of cell proliferation [2]. In this study with the aim to relate the antiproliferative action with an antioxidant activity, were studied: the antiproliferative action, by tritiated thymidine uptake, cell viability, by MTT assay (on BW 5147) and the catalase (CAT), superoxide dismutase (SOD) and peroxidase (PER) activities, by spectrophotometry assays of DME of Tilia x viridis and of the identified compounds (by HPLC and gas chromatography-mass (GC-M)). Results: Mean± SEM of three experiments made by triplicate: Phytochemical composition (retention time min (RT): limonene (L): 7.37±0.5; a pinene (aP): 4.8 ±0.2; â pinene (â P): 5.77±0.3; Scopoletin (SC): 9.5±0.1. Antioxidant activity (U/ml). DME:  CAT: 0.1 µg/ml: 0.450 ± 0.01; 1 µg/ml: 5.8± 0.25; 10 µg/ml: 1.77 ± 0.015; 100 µg/ml: 9.6 ± 0.5; SOD: 0.1 µg/ml: 9 ± 0.5; 1 µg/ml: 30± 2; 10 µg/ml: 35 ± 2; 100 µg/ml: 40 ± 1; 1000 µg/ml: 60 ±1.5. PER (U/mlx10-4): 1 µg/ml: none activity; 10 µg/ml: none activity; 50 µg/ml: 1± 0.1; 100 µg/ml: 1.17±0.1; 500 µg/ml: 1.50±0.15; 1000 µg/ml: 1.30±0.1. The compounds presented CAT,SOD and PER activities (FigsA,B). Antiproliferative action (EC50(µg/ml): DME: 4.70±0.2; L: 35± 2; aP:1100 ±10; â P:114.8± 8; SC: 251±15(FigC) Cell viability (% of basal): DME: 5 µg/ml: 89±5; 100 µg/ml: 80±4; 1000 µg/ml: 6±0.2; L: 10 µg/ml: 98±5; 40 µg/ml: 6.6 ± 0.4; 100 µg/ml: 1.3±0.1;  aP: 10 µg/ml: 90±5; 40 µg/ml: 30± 2; 100 µg/ml: 20± 1; âP: 10 µg/ml: 98± 5; 40 µg/ml: 12±1; 100 µg/ml: 8±0.5; SC: 10 µg/ml: 99±6; 50 µg/ml: 98±4; 100 µg/ml: 99± 7; 1000µg/ml: 68±5; 2000 µg/ml: 38±3. The antiproliferative activity and the decrease in cell viability of DME could be related to the presence of cumarins and monoterpenes and to the induction of antioxidant activity involved principally in the elimination of O2-.
