However, the mechanisms underlying remain unclear andeven less whether obesity affects them. Using cafeteriadiet-induced obesity as animal model, we found that obesity causes aberrantuterine fetal distribution and macrosomia on gestational day (gd) 18.5. Toelucidate whether this aberrant distribution is consequence of fetalreabsorptions or due to alterations in the foregoing embryo spacing, weanalyzed the post implantation loss rates(PIL)andembryo positions just after (gd5.5)and before implantation (gd4.5).β2AR expression (qPCR and westernblot) and localization (immunofluorescence) was analyzed on 4.5 gd. PIL was similar in control and obese rats;however aberrant uterine embryo distribution was detected on gd5.5 and 4.5 inobese animals, indicatingthatobesity alters embryo positioning before implantation. On gd4.5, thetotal number of embryos (p<0.001)and the percentage of blastocysts(p<0.001) were lower in obese rats than controls, while the percentage ofmorulae (p<0.001) was increased in obese animals. ARβ2 localization wassimilar in control and obese rats, showingintense label at myometrium and weak expression at epithelium. ARβ2gene and protein expressions were up regulated in uterus from obeserats(p<0.05 and p<0.001, respectively). Alterations inthe β2ARsignaling beforeimplantation may be one of the mechanisms underlying the aberrant uterineembryo distribution observed in obese animals. Treatments that reverts thisalteration may be useful to prevent implantatory problems and the consequent adverse effects on pregnancy outcome described inobesity.
