Cocaine it is a highly dangerous illegal stimulant that can cause dependence, addiction and long-term consequences when abused. Caffeine is a stimulant drug found in a range of commercially available products such as energy drinks. In addition, in South America there is an endemic use of a drug known as ?Paco? in which it was found quantities of caffeine mixed with other stimulants, including cocaine. Our group has previously described that acute cocaine alters thalamocortical dynamics. The objective of this study was to assess the effects of chronic caffeine/cocaine binge administration on synaptic properties of thalamocortical neurons. Male C57/BL-6 mice were treated with a Coc (10 mg/kg), Caf (5mg/kg), their combination (CC5) (10 mg/kg Coc + 5 mg/kg Caf) or vehicle (Veh), in two protocols, the first one, ?Acute Binge? consisting in 3 ip injections 1 h apart+24 hs, the second one ?Chronic Binge? is an intermittent binge protocol :3 ip injections, 1 h apart, one day on/off for 13 days. We investigated the effect of these treatments on total calcium current and glutamatergic/ GABAergic evoked and spontaneous activity, in Ventrobasal (VB) thalamic neurons.We performed whole-cell patch clamp recordings in brain slices from mice treated with the protocols previously described. We observed that peak calcium current density (pA/pF) mediated by P/Q type calcium channels during whole-cell patch clamp recording of VB neurons decreased significantly at -20mV after Coc+Caf combination (CC5) was administrated .
