Resumen:
Murine perigestational alcohol ingestion until day 10 of gestation (GD) increases resorption at GD7-8. We postulate a link between changes in MMP-2 and -9 expression and cellular alterations on GD8-implantation sites (IS). Ethanol/water-10% was administered to CF-1 females 17 days before and up to GD8 (treated females - TF). Control Females (CF) received only water. Embryo differentiation stage and, histopathology, apoptotic index, collagen-IV and MMP-2 and -9 immunoexpression were evaluated in decidua (De), ectoplacental cone (EC), trophoblast-decidual interface (TDInt), embryo and extraembryonic tissues. TF showed 24h of differentiation delay. Non-delayed-TF-De had decreased collagen-IV, altered MMP-2 and -9 expression and increased TUNEL+ cells. TF-EC-proliferative area was reduced and MMP-9 immunostaining increased. TF-TDIn showed histological abnormalities and increased erythrocyte infiltration, apoptosis and MMP-9 immunoexpression. TF-embryos (66%) showed abnormal neuroectoderm with strong MMP-2 and -9 staining. Periconceptional alcohol exposure changes the morphology of the implantation sites and increases the rates of cell death and collagen-IV, MMP-2 and -9 expression at GD8, suggesting the tissues of the maternal fetal interface are highly susceptible to this treatment, which may contribute with  early embryonic losses.
