We have previously shown the role of 20-HETE, the product of 20-hydroxylation of arachidonic acid, in sustaining the viability of androgen- dependent prostate cancer cells (LNCaP) by acting as a signaling molecule downstream the androgen receptor (AR). Recently, the GPR75 receptor has been identified as the target for 20-HETE. The aim of this study was to identify the role of the 20-HETE/GPR75 axis in the AR transcriptional activity (ARTA) in LNCaP cells. The following methods were used: western blot (expression of GPR75); immunofluorescence (intracellular localization of AR); reporter gene assay (ARTA in LNCaP cells transfected with PSA-Luc or with an empty vector); chemiluminescence (prostate specific antigen (PSA) quantification). Results were analyzed using two tails t-student test, one-way ANOVA followed by Dunnet?s, or two ways ANOVA followed by Sidak?s.The expression of GPR75 was first confirmed in LNCaP cells.
