Cobamides (Cba) are complete corrinoids, namely, a corrinoid containing the a-axial ligand; the name of the Cba depends on the nature of this lower ligand. Cobalamin is a cobamide that have the nucleotide 5, 6-dimethylbenzimidazole as the lower ligand. Previous studies in our laboratory showed that Lactobacillus (Lb) reuteri CRL 1098 produces cobalamin (Taranto y col. 2003). Moreover, the evaluation of the production of cobalamin-like compounds among LAB strains showed that the cell extract (CE) of Lb. curvatus CRL 1000, Lb. coryniformis CRL 1001 and Lb. murinus CRL 1104 strains, was able to correct the requirements of this compound in Lb. delbrueckii subsp. lactis ATCC 7830 (auxothrophic for vitamin B12 ) and in the cobalamin-independent methionine synthase mutants Salmonella enterica serovar Typhimurium AR2680 (metE cbiB) and Escherichia coli 113 (metE). Based on these findings the aim of this work was to isolate and characterized the nature of the cobamides produced by these LAB strains. Each CE was purified using a C18 solid-phase extraction column and by reverse-phase (RP)-HPLC with a diode array detector. The HPLC chromatogram of the standard Cianocobalamin (CN-Cbl) showed a peak with a retention time (RT) of 40,5 min. All the strains assayed showed peaks with a RT similar to the standard CN-Cbl. In addition, the peaks that eluted within 35 min and 45 min were collected and analyzed for their capacity to restore the growth of the indicator strain [Salmonella enterica serovar Typhimurium AR2680 (metE cbiB)]. Only two peaks with a RT of 38 min and 40,5 min from CE of Lb. curvatus, Lb. coryniformis and Lb. murinus showed the same complementation property found in their crude CE. In order to explore the nature of these cobalamin-like compounds, we performed an enzyme immunoassay (ELISA) using specific antibodies against vitamin B12. All the peaks isolated previously were positive for this assay being those with a RT of 40,5 min which showed the strongest reaction. An exception was the peak with RT of 38 min. of Lb. coryniformis. According to the manufacturer (BioPharm), this immunoassay presents a specificity of a 100% for vitamin B12 and 80% for hidroxycobalamin. According to these results, we conclude that Lb. curvatus CRL 1000, Lb. coryniformis CRL 1001 and Lb. murinus CRL 1104 would produce standard cobalamin as well as a vitamin B12 analogue.
