In vitro characterizing of immune responds to these bacteria may give an indication of how these events occur in vivo. We studied if disruption of lipid rafts could influence the effect of five LAB strains on cytokines production by peripheral blood mononuclear cells (PBMCs). Untreated (control) and disrupted-rafts PBMCs (dr-PBMCs) by MβCD teatment were co-incubated with bacterial strains at 37ºC for 4 hours. TNF-α and IL-10 were detected in culture supernatants by ELISA. The results show that L. acidophilus CRL1014, L. rhamnosus CRL1505 and L. reuteri CRL1101 have a similar stimulating effect on TNF-α release by control cells (30 times base line value) and this effect is decreased in dr-PBMCs by 42% of the control cells value. L. casei CRL75 and S. thermophilus CRL1190 has a lower TNF-α stimulating activity and it was reduced in dr-PBMCs by 30% and 50% respectively. L. reuteri strain stimulates 20 times baseline value of IL-10 secretion by control cells, also reduced by dr-PBMCs treatment (12%). Other strains show a similar pattern of stimulation of IL-10 (3 to 5 times baseline value) with small differences between control and dr-PBMCs. Studies are currently in progress to further define the rafts role.
