Medicina-UNT,4Fac de Bioqca,Qca y Fcia-UNT.mmedina@cerela.org.ar Development of vaccines using pneumococcal antigenic proteins is a new strategies in the fight against S. pneumoniae.Objective:To evaluate cytokine production and quantify T and B lymphocytes in lung and peripheral blood (B) of mice nasally immunized with pneumococcal Ag (PppA)+a probiotic (L.casei:Lc).Three successive doses of PppA+Lc (109 cells/mouse) were used and also groups of animals immunized with Lc, PppA and PBS (Control:C) were included. On d0 and 42 after the 1º nasal immunization (NI), samples were harvested. MM:1) Quantify of LT(CD3+,CD4+,CD8+) and LB(IgA+,IgG+) in lung (L) and blood (B) by flow cytometry;2)Cytokines in bronchoalveolar(BAL) and serum(S):IL-4,INF-g e IL-17(ELISA).The total LT did not modified with any of the treatments but PppA+Lc increased the LTCD4+ and decreased the LTCD8+. PppA+Lc increased IgG+cells in S(p <0.05) and the IgA+cells in L. Also,PppA+Lc increased cytokines levels in S and BAL (BAL:d42:IL-4=C=65±7,PppA+Lc=358±26,INF-γ:C=70±8,PppA+Lc=319 ±19).Vaccination with PppA+Lc induced activation of T in S and BAL, evidenced by increased production of cytokines.The IgG+cells in S and IgA+ cells in BAL were increased. Stimulation of humoral and cellular immune response PppA+Lc is importance for the development of a vaccine against pneumococci using as adjuvant, a probiotic strain
