Ethnopharmacological relevance: The aerialpartsof Lippia integrifolia (Gris.) Hieronymus(Verbenaceae),known as incayuyo, reusedbythepeasantpopulationofArgentinafortreatmentofdiseasesrelatedtoa gastrointestinalsystem,mainlyforinflammation ofthestomachandhavealsobeenincludedintothe Argentina FoodCode.Thisstudyaimedtoinvestigatethephytochemicalprofile ofhydrophilicextracts from theherbalmaterialbyLCMS andtoevaluatepotentialpharmacologicalmechanismsrationalizing the traditionaluseof incayuyo aqueousextractspotentialanti-inflammatorytreatmentofgastrointest-inal disorders. Materials andmethods: Phytochemicalprofiling: LCMS fanaqueousdecoction.Antiadhesiveeffects against Helicobacter pylori: in vitro FACSassayusingFITC-labeledbacteriaandAGShumanstomachcells. Influence ofextractsonstomachcellsandRAW264.7macrophages:MTTviabilityassayandBrdU proliferationELISA.Influence ofextractsonIL-6andIL-8secretionfromstomachcellswasquantitatedby ELISA afterinfectionofthecellswith Helicobacter pylori. Influence oftestextractsonmacrophages:phagocytosisofFITC-labeledZymosanparticlesandNOproduction.Antioxidativecapacity:DPPHassay and O2-induced carotenoxidation. Results: LCMS profiling indicatedthepresenceofcompounds 120 with flavonoidhexosides, phenylethanoides(acteoside,isoacteoside)andsesquiterpenes[(epi)lippidulcine,peroxylippidulcine]in hedecoctionextractandsubfractionPhF.Theextractexhibitsstrong in vitro antioxidativecapacity and inhibitedadhesionof Helicobacter pylori to stomachcellsupto40%,whileanEtOH-solublefraction showedinhibitionratesofupto60%.DecoctionincreasedthecellularviabilityofAGScellssignificantly at 410 μg/mL, whileproliferationratewasnotinfluenced. Helicobacter pylori induced IL-8secretionwas significantly reducedbycoincubationofAGScellswiththeextracts.Aqueousextractsstimulated phagocytosisrateofmacrophagesandinhibitedtheLPS-inducedNO-secretion. Conclusions: The traditionaluseofaqueousextractsfrom Lippia integrifolia for gastricinflammation seems toberationalized:besidesanti-inflammatory effectsonstomachcellsantiadhesiveproperties of the extractsagainstthemainbacterialinductorofgastritis Helicobacter pylori wereidentified. Additional effects forstimulationofinnateimmunityandpotentialradicalscavengingeffectsmayadditionally contribute totheactivityoftheextracts.
