Resumen:
The hybrid pine is an economically important alternative for forest plantation in the northeast of Argentine. However, some problems associated with the production of either F1 or F2 seed production limits the rapid expansion of cultivation areas. Tissue culture methodology was successfully applied to find the combination of seed stratification, basal medium and 6-benzyladenine (BA) concentration for inducing the largest number of shoots from regenerative embryos explants. Vegetative buds were originated through a direct pathway from mature zygotic embryos stratified at 4 °C (for 1 to 6 days) and cultured on Murashige and Skoog (either half full strength) semisolid medium containing BA 0.1-5 mg∙L-1. The cultures were incubated in a growth room at 27±2°C with a 14-h photoperiod (180 μmol∙m−2∙s−1PPFD, from fluorescent lamps) for 30 days.The regeneration frequency (93±2.5%) and the number of buds formed per responsive explants (13,2±8,2) was greater with explants stratified at 4 °C for 3 days and cultured on Murashige and Skoog half strength medium plus BA 3 mg·L-1. In general, both parameters diminished due to a higher concentration of either the basal medium or BA.These methodologies can be used in breeding programs for the propagation and cryopreservation of germplasms.
