VCchrysin: a new vanadyl complex with a flavonoid. Anti-tumoral activities in osteoblast-like cells

Resumen:
The family of polyphenolic flavonoids presents estrogenic, anti-allergenic, anti-viral, anti-inflammatory and anti-tumoral effects. In particular, Chrysin is an interesting member of this family, with potent anti-proliferative effects against various malignant cells. Bioavailability of these substances is improved by complexation with organic compounds or by metal coordination. Oxovanadium(IV) compounds show interesting pharmacological activity mainly as insulin enhancers, osteogenic and anti-tumoral agents. In this work we report the biological effects of a interesting complex of vanadyl (IV) with Chrysin ([VO(chrysin)2EtOH]2 on three osteoblastic cell lines. The culture model used involves MC3T3-E1 cells (nontransformed osteblasts derived from mouse calvaria), UMR 106 cells (osteosarcoma derived from rat) and MG-63 (osteosarcoma derived from human bone). The effects on cell viability (crystal violet bioassay, MTT bioassay and neutral red uptake) as well as on cell morphology (DAPI/CM staining and fluorescent microscopy), were investigated. Besides, we studied the mechanisms of action involved in the cytotoxicity (oxidative stress, ratio GSH/GSSH, Arrest of a Cell Cycle and Apoptosis) Vochrysin caused an inhibitory effect on cell viability in the range of 25-100 µM Moreover, the complex increased the level of Reactive Oxigen Species (ROS) over 300% the basal and a decreased a ratio GSH/GSSG at 25-100 µM. (p<0.001). Besides, Vochrysin arrested of a cell cycle in Phase G2 at 6 hs and then sended a cells of apoptosis (evaluated by peak Sub G1) Taken together, these results suggest that Vochrysin is potentially good candidate for future use in alternative anti-tumor trea
