Functional antibodies against the electrogenic Na+/ HCO3-: future therapeutic tool?   Na+/HCO3- cotransport (NBC) regulates the intracelular pH. There are two isoforms of NBC in heart, the electrogenic (NBC1) and the electroneutral (NBC3). In this work, we generated two functional antibodies against extracellular loop domains (Acd3 y Acd4) from heart NBC1. We used isolate cat ventricular myocytes. The antibodies recognize the NBC1 in western blot and immunohistochemistry assay . The total NBC activity was measured in single myocytes with an epifluorescence system using an ammonium pulse (is calculated the net H+ efflux Jh at pH 6.8), and the specific activity of NBC1 using a pulse of potassium for cellular despolarization (DpHi).
