Novel solid-phase strategies towards the generation of libraries of b-lactam compounds

We are developing the solid-phase synthesis of â-lactam compounds with high molecular diversity through organometallic chemistry. Particularly, we have developed the synthesis of cholesterol absorption inhibitor analogues using cross metathesis. Thus, polymer-bound 3-vinyl-b-lactams were synthesized according to a protocol developed by us. Besides, we have developed an efficient protocol for the solid-phase synthesis of 1,3,4-trisubstituted-b-lactams, being the key step a Staudinger reaction using a polymer-bound ketene.
