Resumen:
Excitation Emission Fluorescence Matrix (EEFM)data can be obtained by registering emission spectra at various excitationwavelengths. In order to generate an EEFM, a proper emission spectral range isusually selected, including the wavelengths where the sample constituents areknown to emit. Generally, emission data are registered every 0.5?2 nm, and themain idea is to obtain spectra containing the whole band for the analyte (ca.50-100 nm wide). On the other hand, the number of excitation wavelengths to beused should be selected gathering at least 10-20 data points. The steps aremanually selected, thus if 20 data points are needed for defining a spectrum100 nm wide, then the step should be of at most 5 nm. It should be consideredthat increasing the number of data points in the excitation mode by reducingthe step will unavoidably lead to more experimental time in collecting anentire data matrix, and a reasonable trade-off between working time andexcitation resolution is desirable.
