Resumen:
Nanotechnology has been growing fast as a new area for oral delivery of moleculeswith biological interest. Common drugs from the market usually shownumerous side effects in the organism and a relatively poor biodisponibility. Inthis sense, smart nanodevices encapsulating drugs provide an alternative toavoid the barriers that affect the stability of oral delivery formulations, such asthe extremely acidic pH of the stomach and the presence of gastrointestinalproteases. These systems are combining the benefits of the controlled releaseformulations with the advantages of nanotechnology, such as high surface areato volume ratio, capability to access the deepest interstices of many organs (e.g. intestine), or to penetrate the cell membrane. In the present chapter, the development and application of smart nanopreparations, with growing attention inthe pharmaceutical industry, are discussed. Among them carriers like liposomesare described, seeing how they evolved to solid lipid nanoparticles (SLNs) and, how these evolved to nanostructures of solid lipid carriers. Nanocrystals of pure drugs and of enzymes, another interesting nanoproduct, have emerged as feasible alternatives to overcome the drawbacks of oral administration drugs.More importantly, the use of biopolymers and compatible synthetic polymers become a new tool for the development of nanosystems. Finally, some inorganicmatrices that have been tested in living organisms are considered.
