Background correction is an important step that has to be performed before peak detection and quantitation. When successful, this step greatly simplifies quantitation procedures and enhances accuracy. In fast, online, comprehensive two-dimensional liquid chromatography (LCxLC) coupled with a diode-array detector (DAD) the change in the refractive index of the mobile phase in a very fast gradient causes extremely serious baseline disturbances. The method reported here, can be combined with many of the existing baseline correction methods for one-dimension LC in two dimensional (2D) LC.
