Resumen:
We previously demonstrated that dexamethasone (DXM) increasesadipogenic capacity and number of adipocyte precursorcells (APCs) from retroperitoneal adipose tissue (RPAT). We nowevaluated whether mineralocorticoid (MR) and/or glucocorticoidreceptors (GR) mediate/s these DXM effects. For this aim, stromalvascular fraction cells isolated from RPAT from adult male rats,were cultured up to confluence and maintained in basal culturemedium (CTR) or with DXM either alone (0.25 μM, DXM) orcombined with a GR inhibitor (RU486 1μM, DXM-RU), or a MRinhibitor (Spironolactone 10μM, DXM-SP) or both (DXM-RU-SP)for 48 h. The expression of APCs competency markers (PPAR-γ2and Zfp423), MR and GR were quantified (qPCR). Additionally,cell differentiation was induced and on differentiation day (Dd) 4,we determined the mRNA levels of PPAR-γ2 and C/EBPα (qPCR),and the percentage of PPAR-γ positive cells and its cell nuclearsignal intensity (immunofluorescence). Cell lipid content (Oil-RedO) was measured on Dd 10. We found an increase in PPAR-γ2expression in DXM treated APCs (P
