CD8 T cells are resistant to steroids(the preferred therapy for asthma), and a subset of these cells canproduce type 2 cytokines (IL-5, IL-13) that perpetuate the immuneresponse. Our group has previously shown in a mouse model ofasthma that the instillation of histamine treated DCs exacerbatesTh2/Tc2 type immune response, and that this can be partially revertedwith the use of the H3R/H4R antagonist Thioperamide. Inthe present work we focus on the role of H4R in this effect, usinga highly specific H4R antagonist, JNJ7777120 (JNJ). We generatedthe murine OVA allergy model using two intramuscular doses ofOVA/AlOH (concentración) in a sensitization phase, and 5 consecutivedaily intranasal OVA 3% instillation as a challenge. On day 1of the challenge, mice were also intranasally inoculated with bonemarrow derived DCs (BMDCs) treated or not with HIS (10-6M) andJNJ7777120 (10-5M). Two weeks after day 1 of challenge, mice weresacrificed and the lungs obtained, processed and analyzed. Wefound that, whereas no significant differences were found in IL-13production, another Th2/Tc2 type cytokine, such as IL-5, was which was supplied continuously until weaning of their offspring, includingpregnancy and lactation. Male offspring from rats fed standard(OSD) or cafeteria diet (OCD) were fed with a standard diet, inspectedperiodically, and euthanized at 60 days of age. In the germcells we examined the presence of the reactive oxygen species byflow cytometry using a fluorescent probe (2,7-dichlorofluorescein diacetate),DNA fragmentation by TUNEL kit, mitochondrial functionusing the probe 3.3-diaminobenzidine, the membrane functionalstatus by hypoosmotic swelling test, and the presence of abnormalchromosomes by cytogenetic assay (Evan test).
