However, only a few studies have dealt with the gene expression profile induced by dopamine agonists, and none have provided comparisons with that induced by levodopa. In order to compare the striatal gene expression profile induced by levodopa or pramipexole, three independent groups of male Wistar rats with a partial intrastriatal 6-hydroxydopamine lesion and with readily identifiable motor deficiencies were randomly assigned to 3 groups and received during 3 weeks: water, levodopa 170 mg/kg, or pramipexole 3,5 mg/kg. A fourth group of normal rats was treated with water as a control. Drugs were dissolved in the animal’s drinking water being their only source of liquid. The doses employed were chosen to be capable of causing comparable reversion of contralateral forepaw akinesia in the cylinder test during the third week of treatment. After decapitation, the lesioned striata was dissected and extraction and purification of the total RNA was performed. We used the GeneChip® Gene 1.0 ST array system (Affymetrix) covering all known rat genomic transcripts. After statistical manipulations and validation of the results we hope to identify transcripts related to the therapeutic effects, as well as those possibly related to neuroprotection, plasticity and dyskinesias. We will also hope to be able to speculate on the possible underlying mechanisms and their relationship with the pharmacodynamic and pharmacokinetic properties of the drugs under study.
