PLASTICITY OF MESENCHYMAL STEM CELLS (MSCs) FROM BONE MARROW (BM) OF PATIENTS WITH LUNG AND BREAST CANCER (LCP and BCP)Hofer Erica L, Fernández Vallone Valeria B, Labovsky Vivian, Feldman Leonardo, Bordenave Raúl H, LaRussa Vincent, Chasseing Norma A.MSCs are able to self-renewal and differentiate. Previously, we observed a decrease of the cloning efficiency of MSC to give CFU-F, and of the CFU-F size in BM of untreated advanced BCP and LCP without bone metastasis vs healthy volunteers (HV). Proliferative activity of MSCs is directly proportional to their multipotentialy. To study the influence of tumor development on the relationship between self-renew and plasticity of MSCs, we evaluated the osteo-chondro-adipogenic differentiation of MSCs on 7BM-chemonaive-advanced-BCP, 7LCP and 7HV. BM-MSCs derived from low density 3rd passage were cultured in osteogenic, adipogenic or chondrogenic mediums.Adipogenic:100%HV-cultures responded achieving morphologies with characteristics of this linage vs 57%BCP and LCP. #adipocytes/Petri-dish:HV=468±215a,b;LCP=98±66a;BCP=4±3b (a=p<0.03;b=p<0.001).Chondrogenic:100%cultures developed chondrogenic pellets. +toluidine blue staining with chondrocyte-like lacunae and aggrecan-rich extracellular matrix (EM) were evident in histological sections, and typeII collagen-rich EM. An increase #chondrocytes/pellet was seen in patients versus HV (LCP; BCP vs. HV=p<0.026 and <0.002). Osteogenic: the %of cultures with colony containing osteogenic cells were: 43(BCP), 0(LCP) and 100(HV). Also patient´s cultures showed a 15±7%(BCP) and 10±3%(LCP) of cells differentiated vs. 24±4%(HV). We characterized the cells different to fibroblasts into a pattern of differentiation from grade 0 to 3, 0=fibroblast like, 1=in the way to osteoblast, 2=osteoblast, 3=osteocyte. 43%HV-cultures achieved grade 1 and 57% grade 2-3. In BCP, 57% of the cultures differentiate. From these ones the 75% reached grade 1 and 25% grade 2-3. In 100%LCP-cultures responded to osteogenic medium but grade 1 was achieved by 6/7 LCP-cultures and the other grade 3. Patients showed heterogeneous distribution of calcium as well as an inadequate mineralization of EM.Results showed that patients-BM had a high% of small renewing MSCs, which gave small CFU-F and had less osteo-adipogenic potential. Finally, the in-vitro study of osteogenic differentiation patients-MSCs may be considered as a prognostic factor because its alteration could be the first sign in future bone disease.
