Resumen:
As women increasingly postpone childbearing, many aged females become infertile. Activation of ovarian angiogenesis has emerged as a strategy to prevent ovarian aging. Objectives: 1) Evaluate the local effect of sphingolipid C1P on EPCs mobilization and on ovarian angiogenesis in aged female mice. 2) Analyze the angiogenic potential of follicular fluid (FF) from patients in advanced reproductive age (ARA) in the presence of C1P in endothelial cells (ECs). Aged mice (26-31 weeks) received intraovarian C1P (10µl/ovary; 50µM). Animals were sacrificed 2 and 7 days later. Prior to surgery and sacrifice, peripheral blood (PB) was extracted to evaluate EPCs mobilization by flow cytometry (VE-Cadherin+, Sca-1+). Immunohistochemistry (α-SMA) and Western blot (VEGF-A) were performed on ovaries. ECs were incubated with FF from ARA (38-42 years) and control (
