IMT504, an inmunomodulatory oligodeoxinucleotide, reduces allodynia in an experimental model of inflammatory pain

Chronic pain, either inflammatory or neuropathic, remains a therapeutic challenge. We recently showed that administration of IMT504, a synthetic immunomodulatory oligonucleotide, reduces mechanical allodynia in animals with neuropathic pain. In this study we evaluated the effects of IMT504 administration on the development of allodynia in animals with hindpaw inflammation, induced by intraplantar injection of Complete Freund´s Adjuvant (CFA). Saline-injected and naive rats were used as controls (C). IMT504 administration (20 mg/kg/day, 5 days, sc) was initiated either on the same day of CFA injection or 7 days later (early or late treatment). Mechanical allodynia (painful responses induced by filaments of 6g or less) was assessed using von Frey Test. Animals exposed to CFA developed early mechanical allodynia (p0.05 vs C; days 14-28). Tests using lower IMT504 doses (2 or 0.2 mg/kg/day) resulted in similar anti-allodynic effects. Our study shows that IMT504 treatment both prevents and reduces peripheral inflammation-induced pain, opening a promising therapeutic option.
