ASSOCIATION OF EXPRESSION TO ERBB-2 NUCLEAR WITH KIDNEY CANCER ADVANCED

The renal cellcarcinoma is refractory to chemotherapy andradiotherapy. Themajor subtype of RCC that metastasize are clearcell and papillaryRCC. When it is in advanced stages, decreasedsurvival, and notalways respond to kinase inhibitor therapy. In thiscontext, we exploredthe expression and localization of ErbB-2 andits relationship withthe advance RCC. The techniques used wereimmunohistochemistry,Western Blot and Inmunocytofluorescenceconfocal microscopy.We selected 50 paraffin- embedded tissuesamples from a cohortof archived CCR from the files of the Histolopathologyof Vidal Hospital,Corrientes from 2005 to 2015. Theexperiment in vitrobasal and HRG stimulation they were performedwith cell lines 786-O(RCC) and HEK-293 (Embryonic Kidney). Ourresults show that,the expression to membrane ErbB-2 (MErbB-2)is of 40% (20/50)only Clear cell RCC, are not expression MErbB-2in Oncocytoma andPapilar RCC. However, it is not related to thestage andhistological grade (Chi-square test P=0.8, Stage I vs IIIy IV). We show thatnuclear ErbB2 (NErbB-2) is correlated withhistologic grade(Fuhrman Nuclear Grade, FNG) (Chi-square test**P=0.007 FNG 1 vs 2y 3). Here, we explored ErbB3 expressionbecause we found inthe correlation analysis ErbB3 and ErbB2mRNA expression (TCGARenal datasets in Oncomine ResearchEdition). Our resultsshow relationship between NErbB-3 andNErbB-2, in tissuesamples and in vitro in 786-O cells with HRGstimulation.Furthermore, In vitro experiments confocal showNErbB2 and NErbB3with HRG in 786-O (t-test **P=0.01). Ourfindings showed ErbB-2is implicated in the carcinogenesis renal.Is very important tolocalization ErbB-2, because in the clear cellrenal cell carcinoma,ErbB-2 nuclear It is associated with poorprognosis. Ourchallenge is elucidate the molecular mechanismsof partner ErbB-2/ErbB-3 in renal cellcarcinoma.
