Ovarian tissue cryopreservation allows storage of large numbers of oocytes containedin preantral follicles (PAF).The aim of the present study was to perform a quantitative analysis of the effect of ethylene glycol (E), DMSO (D) and a combination of both (Mix), after exposition (toxicity) or vitrification, in the preservation of porcine PAF histological structure.Fragments of ovarian cortex were exposed to the vitrification solution (TCM 199-Hepes;30% E, 30%D or 15%E and 15% D; 20% fetal bovine serum; 0.25M sucrose). Samples for the vitrification test were then vitrified in cryotubes and stored in liquid nitrogen. Quantitativeanalysis was performed on digital photographs of samples stained with haematoxylin-eosin.Nuclear (NA), cytoplasmic (CA) and total area (AT) of each follicle were obtained usingthe Leica Qwin Plus® software, and then the NA/CA ratio was calculated. In primordial follicles, a significant reduction of all the morphometric parameters was found after the toxicity test. Vitrification process produced a more pronounced reduction in these parameters.The best vitrification protocol in our working conditions proved to be TCM 199-HEPESwith ethylene glycol 30%, and 0.25 M sucrose.
