The Endocannabinoid Anandamide regulates Oxytocin receptor in human placenta

Growing evidence indicates that endocannabinoids are involved in regulating theinitiation of labour. Hence wehypothesize that Anandamide (AEA),one of the main endocannabinoids, modulates oxytocin signaling in humanplacenta at term. The objectives were: 1) to study the expression of the endocannabinoidsystem in human placenta from vaginaldelivery (VD) or cesarean section (CS) at term, and 2) to analyze the effect of AEA onoxytocin receptor (OTR) expression in placenta. To this purpose, placentas fromVD or CS were used for RT-qPCR, Western Blot, enzymatic activity andELISA assays. In accordance with the increase in AEA serum levels observed inlabouring women, we found a significant (p<0.05) decrease in the proteinlevels and enzymatic activity of FAAH, the enzyme that degrades AEA, in VDplacentas. We found no differences in CB1 protein content between VD and CSplacentas, while TRPV-1 protein levels were significantly (p<0.05) lower inVD placentas. When analyzing oxytocin and OTR, we verified that the placentaproduces oxytocin and that VD placentas present a significantly (p<0.05) higherOTR mRNA levels compared to CS placentas. Thereby, explants from CS placentaswere cultured in the presence of different concentrations ofR-(+)-Methanandamide (Met-AEA), a stable AEA analogous, for 3, 6 or 20 hours.We observed that Met-AEA significantly (p<0.05) increases OTR mRNA levelsafter 20 hours of culture. To analyze whether Met-AEA acts on either thecytotrophoblast or the syncytiotrophoblast, we cultured and differentiated BeWocells in the presence or absence of Met-AEA for 48 hours. We observed that Met-AEAsignificantly (p<0.05) increases OTR expression in the syncytiotrophoblastcells, while no effect was observed on the cytotrophoblast. Our findingsindicate a differential expression of the endocannabinoid system between VD andCS placentas, and suggest that AEA stimulatesOTR expression in the placenta at term.
