Changes in the oocyte integrity and bone marrow induced by 3-methylcholanthrene and prevented by α-naphthoflavone

Resumen:
3-Methylcholanthrene (3MC), a polyaromatic hydrocarbon, is an environmental pollutant that causes reproductive toxicity and genotoxicity. Previous studies showed that 3MC alters the ovarian function by affecting the follicle integrity in rodents, and causes DNA damage in peripheral blood, bone marrow and oocytes. Now, we studied the effect of daily exposure to 3MC on the oocyte integrity, induction of chromosome aberrations in bone marrow and the ability of α-naphthoflavone (αNF) to prevent this action. To this end, immature rats were daily injected with a combination of 3MC (0.1 and 1 mg/kg) and αNF (80 mg/kg) for 20 days. Oocyte integrity, by morphological parameters; oocyte viability, by hoechst 33342 and propidium iodide; and chromosome aberrations (CA) in bone marrows cells were evaluated. All statistical analysis was performed by two-way ANOVA with Bonferroni post-test. Levene?s test and a modified Shapiro-Wilk test were used to assess homogeneity of variances and normal distribution, respectively.
