Remodelling thespiral arteries by the trophoblast is crucial during human implantation.Previously, we observed that lysophosphatidic acid (LPA), a small and bioactivephospholipid, enhances vascularization of first trimester trophoblast cells byincreasing tubulogenesis through LPA3 (an LPA receptor). Estradiol (E2) andprogesterone (P4) are the master hormones that orchestrate the events that takeplace during implantation. Our objective was to study if E2 and P4 regulate theaction of LPA/LPA3 in vascularization of trophoblasts. First trimestertrophoblast cells (HTR-8/SVneo line) were seeded on a reduced growth factorbasement membrane matrix (Geltrex®), incubated with E2 (10-8, 10-9and 10-10 M) or P4 (10-6, 10-7 and 10-8M) in the presence of a selective LPA3 antagonist (DGPP 10-4 M) or anon-selective LPA3 antagonist (8 Br-LPA 5x10-6 M), and assayed for tubeformation during 6h. 8 Br-LPA antagonizes LPA1, 2, 3 and 4 receptors.
