EL ESTRADIOL Y LA PROGESTERONA REGULAN LA VASCULARIZACIÓN DEL TROFOBLASTO A TRAVÉS DEL SISTEMA LPA/LPA3.

Remodelling ofthe spiral arteries by the trophoblast is crucial during human implantation.Previously, we observed that lysophosphatidic acid (LPA), a small and bioactivephospholipid, enhances the vascularization of first trimester trophoblast cellsby increasing tubulogenesis through LPA3 (an LPA receptor). Estradiol (E2) andprogesterone (P4) are master hormones that orchestrate the events that takeplace during implantation. Our objective was to study if E2 and P4 regulate theaction of LPA/LPA3 in vascularization of trophoblasts. Thus, first trimestertrophoblast cells (HTR-8/SVneo line) were seeded on a reduced growth factorbasement membrane matrix (Geltrex®), incubated with E2 (10-8, 10-9and 10-10 M) or P4 (10-6, 10-7 and 10-8M) in the presence of a selective LPA3 antagonist (DGPP 10-4 M) or anon-selective LPA3 antagonist (8 Br-LPA 5x10-6 M), and assayed for tubeformation during 6h. 8 Br-LPA antagonizes LPA1, 2, 3 and 4 receptors. Weobserved that the incubation with E2 (10-8 M) or with P4 (10-7M) increased tube formation (p<0.05) of HTR-8/SVneo cells. While theco-incubation of E2 with DGPP partially reversed tubulogenesis, the treatmentwith E2 + 8 Br-LPA completely blocked the effect. Both DGPP and 8 Br-LPAdecreased P4 stimulation to control level. Our results suggest that E2 and P4regulate the vascularization of first trimester trophoblasts through LPA and atleast LPA3 receptor.
