Background: We reported the presence of autoantibodies (autoAbs) in the immunoglobulin (Ig) G fraction from breast cancer patients (BCP) sera that promote tumor progression by activating muscarinic acetylcholine receptors (mAChR) in tumor cells. Since this type of autoAbs have been localized in tumor microenvironment exerting a complex network of interactions in which dendritic cells (DC) could be also implicated, we investigated whether autoAbs can modulate the expression of maturation markers and cytokine production by DC through cholinergic activation. Methods: DC were obtained from human monocytes and cultured with lipopolysaccharide during 24h to obtain mature DC (mDC). Then, mDC were treated with IgG from BCP in the absence or presence of cholinergic antagonists (atropine or mecamylamine). The expression of HLA-DR and CD86 were measured by flow cytometry and the production of cytokines in DC culture medium was quantified by ELISA.
