THE OROGASTRIC VACCINATION WITH AN ATTENUATED STRAIN OF SALMONELLA TYPHI, TRIGGERS AN IMMUNE RESPONSE THAT PROMOTES METASTATIC CELLS ELIMINATION IN THE LIVER

Resumen:
Previously, we showed that the administration of CVD 915 via the orogastric route (o.g.) 24 h before tumor challenge, can prevent implantation of LM3 cells in the liver, in a mouse model of liver metastasis. Twenty days after tumor challenge, treatment with CVD 915 significantly decreased the occurrence of liver metastases and total tumor volume. The aim of this work is to elucidate the immune mechanisms involved in the antitumor effect. For this purpose, 1x109UFC of CVD 915 was inoculated via o.g. in a group of BALB / c mice, while another group received PBS as a control. After 24 h, mice were injected with 100000 LM3 cells (mammary adenocarcinomas) into the spleen in a surgical frame and underwent splenectomy. After 24 h of the o.g. gavage with the bacteria, we find an innate immune response activation at the liver, characterized by a significant increase in IFN-γ production (p <0.05) by intrahepatic immune cells (IHIC) from treated mice. Also, we found an increase in the population of natural killer and neutrophils (Gr1 +) in the spleen (p <0.05). Furthermore, we found after 20 days of tumor challenge, a specific immune response in the liver with an increase in the population of CD4 + cells of the IHIC (p <0.05) and a significant increase in the IFN-γ, TNF-α and IL-2 production in cultures of IHIC from treated mice (p <0.05). Indeed, we demonstrated the IFN-γ and TNF-α tumor-specific production when co-culturing the IHIC or the liver draining lymph nodes (the portal and celiac) cells with LM3 cells (p <0.05). These results indicate that immunization with CVD 915 via o.g. route induces an antitumor immune response which reduces the metastases development in a vital organ. The use of this vaccine as an adjunct therapy to conventional cancer treatments, such as surgery, should be assessed in the future.
