Melatonin inhibits lipopolysaccharide-induced preterm delivery and reduces mortality in the newborn mouse

Preterm delivery is the leading cause of neonatal mortality and promotes delayed physical and cognitive development in children. Intra-amniotic infections are one of the main causes of preterm birth (PTB). Melatonin, a potent endogenous antioxidant, inhibits prostaglandins synthesis and uterine contractility. We developed a mice model of inflammation-associated PTB to mimic the clinical consequences of preterm labour. We obtained 100% incidence of PTB administrating LPS on day 15 of pregnancy. Melatonin prevented PTB in 80% of cases and conferred protection to the fetuses. Melatonin also blocked LPS-induced PGF2alpha synthesis, Nitric oxide (NO) production, COX-2 and iNOS protein levels augmentation. Fetuses are born alive and had no gross changes comparing to the control animals. Maternal treatment with LPS and melatonin did not affect newborn mice weight during the first month of life. These results suggest that melatonin could be an useful tool to prevent preterm labour and improve offspring.
