STUDY OF THE ADJUVANT EFFECT OF CLIPTOX ON THE IMMUNE RESPONSE INDUCED BY AN INACTIVATED VACCINE AGAINST FOOT AND MOUTH DISEASE VIRUS AND A DNA VACCINE AGAINST BOVINE HERPES VIRUS-1.

Cliptox TM , is a natural mineral product with adjuvant activity tested with classical antigens. In this research we studied the efficiency of the adjuvant on the immune response induced by inactivated vaccine against Foot and Mouth diseases virus. As well we wanted to test  a genetic vaccine against Bovine Herpes Virus-1 with the same adjuvant. Balb/C mice were vaccinated subcutaneously, or subcutaneously+intranasaly or intranasaly only with: inactivated FMDV, inactivated FMDV and Cliptox  or inactivated FMDV and a commercial adjuvant. The results obtained demonstrate that the specific humoral immune response was significantly increased using Cliptox. The IgG1 and IgG3 were the principal isotype induced by Cliptox similar to the kind of immunity induced by the active virus.   On the other hand we have investigated the use of Cliptox as adjuvant using a DNA vaccine based on the BHV-1 glycoprotein D (gD) in the PCI Neo vector. The antibody response induced in mice by DNA truncated gD (secreted version) formulated with Cliptox showed that the immunomodulator affect the total specific humoral response. The isotypes induced by the adjuvant was type Th1/Th2. The cellular immune response induced in mice immunized with genetic vaccine and the adyuvant has been studied. When total spleen cells were targeted with specific monoclonal antibodies against surface markers, a significant increase in the macrophage cells was observed. These was evidenced in the groups receiving PCI Neo+ Cliptox in comparation with PCI Neo group.  Our results indicate that the incorporation of Cliptox into DNA vaccines against BHV-1 induces an increase of the specific humoral and cellular immune response in mice.
