Resumen:
Electrical stimulation for the treatment of pharmacoresistant epilepsies is now being widely studied both clinically and experimentally. The present study was designed to investigate the effect of unilateral BPS of hipocampal on seizure development induced by hipocampal kindling in rats. BPS was applied inmediately after to kindling stimulation, daily and consisted of 0.5 s trains of biphasic square wave pulses, 0,2 s. in duration, at a frequency of 50 Hz., intensity of 50 ìA - 200 ìA dependent of ADT for each animal, for 40 consecutive days. BPS significantly slowed the progression of behavioral seizure stage and decreased the mean AD duration during kindling acquisition (p<0.0001). This marked BPS-induced decrease in kindling rate was predominantly due to the animals in the BPS groups remaining longer in stages 1 and 3. This protective pattern provides further evidence that BPS may play a crucial role in the progression and secondary generalization of focal seizures. We have not determined what mechanisms contribute to the strong protective effect of BPS. However, the mechanisms proposed to account for these phenomena include inhibitory mechanisms, possibly located in specific cortical layers or cerebral pathways and possibly mediated by outward ionic currents. These findings may provide a new insight into establishing an effective and long-lasting therapy for seizure prone people, especially those with temporal lobe epilepsy in future.
