ANGIOTENSIN-(1-7) INDUCES A DIFFERENTIAL MAS RECEPTOR TRAFFIC IN NEURONS OF SPONTANOUSLY HYPERTENSIVE RAT In hypertension there is an exacerbation of the classic pressor arm of the renin angiotensin system (RAS) in detriment of the depressor arm constituted by angiotensin converting enzyme 2/angiotensin (Ang)(1-7)/Mas receptor (R). Our aim was to evaluate if a differential regulation of the neuronal Mas R contributes to an unbalanced RAS and therefore to the development of hypertension in spontaneously hypertensive rats (SHR). Methods: Primary culture of neurons (N) from hypothalamus and brain stem of SHR and normotensive rats (Wistar Kyoto-WKY) were incubated in the presence or absence of Ang(1-7) (10-6M) during 15 or 30 min. MasR expression was evaluated by western blot (WB) and immunofluorescence in non-stimulated N, and MasR trafficking by colocalization with Lysotracker, a lysosome marker, or Hoescht, a nuclear marker.
