Proliferating Cell Nuclear Antigen (PCNA) is highly expressed in divinding cells. This nuclear protein regulates cell cycle, replication and DNA repair. In neutrophils, PCNA is predominantly expressed in cytoplasm and has been associated to cell survival in this cell type. At the beginning of gestation, endometrial cells undergo a constant rate of proliferation and vascularization in response to multiple factors such us PCNA and Hypoxia-inducible factor (HIF). The goal of this study was to determine the possible interaction between HIF and PCNA in human endometrial cells. To reach this purpose we performed in vitro experiments using the endometrial cell line ECC-1. The cells were treated with CoCl2, a hypoxic mimicking agent, in order to stimulate HIF expression. Proliferation and viability tested by trypan blue determined that CoCl2 significantly increased cell proliferation without afecting viability with respect to controls. In order to determine cellular localization and action of PCNA we studied by western blot the differential expression of nuclear and cytoplasmatic proteins after 6 hs of CoCl2 treatment. Our results demonstrated that PCNA had a highly expression in the cytoplasmatic fraction with respect to controls due to CoCl2 treatment. By immunofluorescence microscopy we confirmed that PCNA is expressed predominantly in the nucleus and translocates to cytoplasm after the treatment with CoCl2. Our results demonstrated that in endometrial cells PCNA is highly expressed during proliferation and translocates to cytoplasm under hypoxic mimicking conditions. Our findings suggest that exists a crosstalk between HIF and PCNA during proliferation in endometrial cells.
